Viabilitas Bacillus S1, SS19 dan DA11 pada Medium yang Terpapar Logam Kromium (Cr)

Abstract


Kromium (Cr) tergolong logam berat yang dapat masuk ke lingkungan melalui limbah industri sehingga mencemari lingkungan. Telah diketahui terdapat bakteri resisten Cr yang diisolasi dari limbah yang mengandung logam Cr. Penelitian ini bertujuan untuk mengetahui resistensi dan viabilitas Bacillus yang tercekam Cr. Uji resistensi Bacillus dilakukan pada medium nutrient agar yang mengandung K2Cr2O7 50 mg/L, 100 mg/L, 150 mg/L. Uji viabilitas Bacillus dilakukan pada 3 isolat yang dipilih dari uji resistensi. Isolat tersebut dikategorikan lebih resisten dari isolat yang lain. Uji viabilitas dilakukan pada media nutrient broth yang mengandung K2Cr2O7 50, 100, dan 150 mg/L. Kepadatan sel diukur dengan spektrofotometer UV-Vis pada panjang gelombang 600 nm selama 24 jam. Hasil penelitian menunjukkan isolat Bacillus resisten pada medium nutrient agar yang mengandung logam Cr pada konsentrasi ≤ 250 mg/L. Pola pertumbuhan isolat Bacillus yang tercekam logam Cr relatif lebih rendah dibandingkan kontrol. Semakin tinggi konsentrasi Cr yang digunakan, semakin rendah pola pertumbuhan Bacillus.
