Resistensi Azotobacter terhadap HgCl2 yang Berpotensi Menghasilkan Enzim Merkuri Reduktase

Abstract


Merkuri adalah logam berat yang sangat tosik dan berbahaya. Beberapa bakteri dapat hidup di habitat yang tercemar merkuri disebut bakteri resisten merkuri, satu diantaranya adalah Azotobacter. Tujuan penelitian adalah mendapatkan isolat unggul Azotobacter dari lahan Eco Urban Farming ITS yang resisten terhadap HgCl2 yang berpotensi menghasilkan enzim merkuri reduktase. Isolasi dilakukan menggunakan media selektif Azotobacter agar. Pengujian resistensi merkuri dilakukan dengan metode goresan agar miring yang megandung HgCl2. Kurva pertumbuhan menggunakan spektrofotometri pada panjang gelombang 600 nm. Hasil isolasi mendapakan 10 isolat Azotobacter resisten HgCl2. Isolat A5, A6 dan A9 mempunyai resistensi tertinggi pada penambahan HgCl2 20 mg/L. Pada A7, A10 dan A. chroococcum resisten mencapai 15 mg/L sedangkan A1a resisten mencapai 10 mg/L. Sedangkan ang hanya resisten sampai 5 mg/L adalah isolat A1b, A2, A3, dan A8. Dari kesepuluh isolat yang berpotensi menghasilkan enzim merkuri reduktase adalah isolat yang memunyai resisten tertinggi yaitu isolat A5 dan A9.
