Adaptasi Isolat Bakteri Aerob Penghasil Gas Hidrogen pada Medium Limbah Organik

Abstract
Hidrogen merupakan salah satu energi alternatif yang efisien dan ramah lingkungan. Hidrogen dapat dihasilan oleh bakteri aerob dengan menggunakan limbah organik sebagai substratnya. Perbedaan medium dari medium pengaya ke medium limbah organik cair akan menyebabkan stres pada bakteri. Sehingga perlu adanya proses adaptasi agar bakteri mampu bertahan hidup dalam medium limbah organik. Pengadaptasian dilakukan dengan memindahkan inokulum dari medium pengaya ke medium limbah organik cair. Setelah berumur 24 jam dipindahkan kembali ke medium limbah organik cair yang baru dan diukur pertumbuhannya dengan optical density (OD600nm) setiap 24 jam hingga 3 kali transfer adaptasi. Pertumbuhan bakteri aerob isolat A6, A27 dan A31 selama adaptasi menunjukkan adanya kenaikan pertumbuhan setelah mengalami tiga kali transfer adaptasi. Isolat bakteri A31 menunjukkan proses adaptasi yang signifikan mulai dari transfer I hingga transfer III. Adaptasi pertama menunjukkan adanya peningkatan OD dari 0.56, 1.02, hingga 1.3. Adaptasi kedua dari 0.62, 2.16. hingga 2.34. Adaptasi ketiga 0.54, 2.3, hingga 2.88. Hal ini menunjukkan bakteri A6, A27 dan A31 mampu hidup dalam medium limbah organik cair
